Signaling begins at the GHRH receptor, a class B G protein-coupled receptor displayed on somatotroph cells of the anterior pituitary. Receptor occupancy activates Gs proteins, which raise adenylyl cyclase activity and intracellular cyclic AMP, in turn driving protein kinase A dependent pathways. The downstream output is synthesis and pulsatile secretion of growth hormone into the bloodstream. Hepatic tissue and peripheral sites respond by increasing insulin-like growth factor 1 production. Somatostatin and IGF-1 itself supply negative feedback that caps the size and duration of each secretory burst.
Metabolic interest in this compound centers on fat distribution rather than on hormone levels alone. Imaging trials in adults with excess abdominal fat report reductions in visceral adipose tissue, while subcutaneous depots change comparatively little. Growth hormone and IGF-1 are presumed to carry the effect, but the separate contribution of each is not firmly established. Whether these changes persist after treatment stops, and whether they alter longer-term health outcomes, remain open questions that published work does not answer consistently.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.

The hexenoyl cap slows the enzyme step that trims the amino terminus of native GHRH, the same step that shortens its active lifetime in circulation. As a result, the modified peptide persists longer in plasma than the unmodified hormone in side-by-side comparison. Receptor activity stays broadly comparable, because the added group sits away from the residues that contact the binding site. This combination, preserved receptor activity with reduced degradation, explains why the analog was developed instead of the native sequence.
Several compounds share the GHRH framework, including sermorelin, the shorter 1-29 fragment, and other analogs built on the full 1-44 chain. Naming follows a common convention: a stem that identifies the peptide plus a suffix marking analog status. Reports may describe tesamorelin by its sequence fragment, as a GHRH(1-44) analog, or by its amino-terminal modification. Indexing the compound therefore requires searching all of these forms, since some older literature predates the current international nonproprietary name.
Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
The native hormone is produced in the hypothalamus and acts on the anterior pituitary. Binding of GHRH to its receptor stimulates synthesis and release of growth hormone into circulation. Because the analogue retains the receptor-binding region of the parent sequence, it engages the same receptor and triggers the same downstream signaling. The result is increased growth hormone secretion from pituitary cells, which in turn influences hepatic production of insulin-like growth factor 1. This axis is the basis for the compound's measured biological effects.
Interest in this peptide developed because native GHRH has a short circulating lifetime. The N-terminal modification slows cleavage by dipeptidyl peptidase IV, an enzyme that removes the first two residues of many peptides and terminates their activity. Slower degradation means a longer window of receptor stimulation per administration. This design logic parallels other modified peptide hormones, where a small chemical change at a vulnerable site yields a more durable molecule without altering the core mechanism of action.
Lead coolant temperature would be around 540 °C, giving a high efficiency of 43%, primary heat production of 700 MWt yielding electrical power of 300 MWe. The operational lifespan of the unit could be 60 years. The design was expected to be completed by NIKIET in 2014 for construction between 2016 and 2020. In January 2026, Rosatom reported that BREST-OD-300 remained under construction and that the metal shell of its central reactor cavity had been installed during 2025.
Machine learning algorithms require large amounts of data. The techniques used to acquire this data have raised concerns about privacy, surveillance and copyright. AI-powered devices and services, such as virtual assistants and IoT products, continuously collect personal information, raising concerns about intrusive data gathering and unauthorised access by third parties. The loss of privacy is further exacerbated by AI's ability to process and combine vast amounts of data, potentially leading to a surveillance society where individual activities are constantly monitored and analysed without adequate safeguards or transparency. Sensitive user data collected may include online activity records, geolocation data, video, or audio. For example, in order to build speech recognition algorithms, Amazon has recorded millions of private conversations and allowed temporary workers to listen to and transcribe some of them. Opinions about this widespread surveillance range from those who see it as a necessary evil to those for whom it is clearly unethical and a violation of the right to privacy. AI developers argue that this is the only way to deliver valuable applications and have developed several techniques that attempt to preserve privacy while still obtaining the data, such as data aggregation, de-identification and differential privacy. Since 2016, some privacy experts, such as Cynthia Dwork, have begun to view privacy in terms of fairness.
A study on Neanderthal teeth from the Chagyrskaya Cave in the Altai Mountains (Russia), indicating that the studied sample overall falls within the known Neanderthal phenotypic variability but also preserves specific morphological traits, is published by Gicqueau et al. (2026). Massilani et al. (2026) present a high-quality genome of an approximately 110,000-years-old Neanderthal individual from the Denisova Cave (Russia), providing evidence of a closer relationship of the studied individual to a 120,000-years-old Neanderthal from the same cave than to a 80,000-years-old individual from the Chagyrskaya Cave or to European Neanderthals, evidence of gene flow from Denisovans in both Neanderthals from the Denisova Cave, and evidence of differentiation between Altai and European Neanderthals comparable to that of the most differentiated populations of modern humans. Zubova et al. (2026) report evidence of deliberate invasive dental caries intervention in a 59,000-years-old Neanderthal molar from the Chagyrskaya Cave. Evidence from the study of rhinocerotid remains from Middle Paleolithic sites in France and Spain, interpreted as consistent with use of rhinoceros teeth as tools (including soft hammers and anvils) by Neanderthals, is presented by Sanz-Royo et al. (2026). Picin et al.
Petroleum in some form has been used since ancient times. More than 4,300 years ago, bitumen was mentioned when the Sumerians used it to make boats. A tablet of the legend of the birth of Sargon of Akkad mentions a basket which was closed by straw and bitumen. More than 4,000 years ago, according to Herodotus and Diodorus Siculus, asphalt (bitumen) was used in the construction of the walls and towers of Babylon; there were oil pits near Ardericca and Babylon and a pitch spring on Zakynthos. In Babylon, petroleum was used for road construction, ship caulking, and medicine. The use of petroleum in ancient China dates back more than 2,000 years. The I Ching, one of the earliest Chinese writings, cites that oil in its raw state, without refining, was first discovered, extracted, and used in China in the 1st century BCE. In addition, the Chinese were the first to record the use of petroleum as fuel as early as the 4th century BCE. By 347 CE, oil was produced from bamboo-drilled wells in China. In the 7th century, petroleum was among the essential ingredients for Greek fire, an incendiary projectile weapon that was used by Byzantine Greeks against Arab ships attacking Constantinople. Crude oil was distilled by Persian chemists, with clear descriptions given in Arabic handbooks such as those of Abu Bakr al-Razi. In the 9th century, oil fields were exploited in the area around modern Baku, Azerbaijan. These fields were described by Abu Bakr al-Razi in the 10th century and by Marco Polo in the 13th century, who describes the output of those wells as hundreds of shiploads.

== 2024 premiership campaign == In January 2024, The PPP published its electoral manifesto for the upcoming general elections the following month, with Bhutto Zardari serving as the party's candidate for prime minister. Bhutto Zardari promised to bring a "transformational change" by addressing several issues, including water and food insecurity, and the country's economic woes. His campaign also relied on youth appeal, with Bhutto Zardari championing the fight against climate change to rally Pakistan's youth. Ahead of the general elections, Bhutto Zardari toured all four provinces in January, with his sister, Aseefa Bhutto Zardari, assisting his campaign. Following the election, which produced a hung parliament, Bhutto Zardari announced his withdrawal from the race and that the PPP would not participate in any post-election federal cabinet. On 13 February, Bhutto Zardari's father, Asif Ali Zardari, who was representing the PPP, announced that a coalition government would be formed, with Shehbaz Sharif as the Prime Minister. Bhutto Zardari added that the PPP would nominate Zardari to become the next president, while it would also field candidates for the Senate chairmanship and the speakership of the National Assembly.

Attached gingiva The attached gingiva represents the apical continuation of the marginal gingiva. It is firm, resilient, and immobile due to its dense connective tissue attachment to the periosteum of the alveolar bone. The width of attached gingiva varies depending on tooth position and plays an important role in maintaining periodontal health.
==== Essential fatty acids ==== There is insufficient evidence to determine that omega-3 fatty acid has any effect on depression. A 2016 review found that if trials with formulations containing mostly eicosapentaenoic acid (EPA) are separated from trials using formulations containing docosahexaenoic acid (DHA), it appeared that EPA may have an effect while DHA may not, but there was insufficient evidence to be sure. A 2020 meta-analysis showed that a high dose of omega-3 polyunsaturated fatty acid (>2 g/day) used as an adjuvent improved depressive symptoms.

=== Human civilization: a model for extrapolation === Kardashev poses the following question: "Is it possible to describe the development of a civilization in general terms over large cosmological periods?" Now many of the fundamental parameters that characterize the development of civilization on Earth are growing exponentially. In the field of energy, astronomer Don Goldsmith estimated that the Earth receives about one billionth of the Sun's energy, and that humans use about one millionth of it. So we consume about one millionth of a billionth of the Sun's total energy. Since human expansion is exponential, we can determine how long it will take for humanity to go from Type II to Type III according to Michio Kaku. Thus, the rate of development of our own world remains the only criterion for extrapolating the state of civilizations older than humanity. The same is true for social values and basic needs according to Ashkenazi. Therefore, the time to double technical knowledge is about 10 years, and to double energy output, available reserves, and population is about 25 years. Two scenarios are then possible: spatial expansion or energy stagnation, the latter being possible only for 125 years, according to Kardashev, using the following relationship
== Stability == The stability of a nanoparticle is a term often used to describe the preservation of a specific, usually size-dependent, property of the particle. It can refer to e.g.: its size, shape, composition, crystalline structure, surface properties or dispersion within a solution. The interfacial layer of a nanoparticle can aid these types of stabilities in different ways. The ligands can bind to the different facets of a nanoparticle, the size and type of which will determine the way the ligands will be ordered. The way the ligands are attached to the particle, ordered disordered or somewhere in between, plays a crucial role in the way different particles will interact. This in turn affects the reactivity of the nanoparticle, which is another way to look at the stability of the particle.
Single malt Scotch whisky must have been distilled at a single distillery as a batch process using a pot still distillation process and made from a mash of 100% malted barley. Single malt means that the whisky has not been blended elsewhere with whisky from other distilleries. A single malt Scotch must be distilled in Scotland and matured in oak casks in Scotland for at least three years, although most single malts are matured longer and also must be bottled in Scotland. Single grain Scotch whisky is a Scotch whisky distilled at a single distillery but, in addition to water and malted barley, may involve whole grains of other malted or unmalted cereals. Grain whisky can be distilled continuously in continuous stills or column stills. Single grain whisky can essentially be seen as any spirit from one distillery which qualifies as whisky but does not qualify as malt whisky. "Single grain" does not mean that only a single type of grain was used to produce the whisky; rather, the adjective "single" refers only to the use of a single distillery (and making a "single grain" generally requires using a mixture of grains, as barley is a type of grain and some malted barley must be used in all Scotch whisky - although a single grain whisky can be made entirely from malted barley and continuously distilled). Excluded from the definition of "single malt Scotch whisky" or "single grain Scotch whisky" is any spirit that qualifies as a blended Scotch whisky.
=== Source, Steam, and Half-Life 2 (2003–2010) === In 2003, Valve moved to Bellevue, Washington, and reincorporated as Valve Corporation. In 2010, the office moved to a larger location in Bellevue. In 2016, Valve signed a nine-floor lease in the Lincoln Square complex in downtown Bellevue, doubling the size of its offices. Valve began developing Half-Life 2 six months after the release of the first Half-Life, using its new in-house engine, Source. With advanced physics systems and an increased focus on story and characters, it received critical acclaim upon its release in 2004. By 2011, it had sold 12 million copies. In 2002, Valve launched Steam, a digital storefront and delivery platform. Steam initially offered only Valve games, and was mandatory to install Half-Life 2, but it later became a publisher of third-party games. As Valve became its own publisher via Steam, it transitioned to a flat organization. Outside of executive management, Valve does not have bosses and uses an open allocation system, allowing employees to move between departments at will. In July 2005, Electronic Arts was announced to partner with Valve for a multi-year game distribution agreement, following Vivendi Universal's promise to end distribution on August 31. After having taken five years to develop Half-Life 2, Valve moved to episodic development, planning to release shorter games more frequently. Half-Life 2: Episode One, the first in a planned trilogy of episodic Half-Life 2 sequels, was released in 2006.

The amino acid sequence matches human growth hormone-releasing hormone, but the amino terminus carries a trans-3-hexenoyl group instead of a free amine. That single structural change chiefly affects enzymatic stability rather than receptor selectivity.
Growth hormone released from the pituitary stimulates IGF-1 production in the liver and other tissues, so circulating IGF-1 generally rises during exposure. The size of the rise varies between individuals and depends on baseline hormonal status and other concurrent factors.
Reported imaging studies focus on visceral adipose tissue, where reductions are more consistently observed across trials. Subcutaneous depots show smaller and less reproducible changes, so the two compartments should not be treated as equivalent.
It is a synthetic peptide analog of human growth hormone-releasing hormone. It is used clinically to reduce excess visceral abdominal fat in adults with HIV-associated lipodystrophy. It works by stimulating pituitary growth hormone release.
