=== Expert panel investigation === During Letby's 2022–2023 trial, the prosecution's expert witness, Dewi Evans, relied on a 1989 article by Canadian neonatologist Shoo Lee to support a diagnosis of air embolism as the mechanism of death in several cases. Lee did not learn that his research had been cited in the trial until after the verdict. He expressed concern that his work had been misapplied. Following Letby's unsuccessful application for leave to appeal in 2024, at the instruction of Letby's defence, Lee convened a panel of 14 medical experts to "review all of the cases from the trials." Letby's defence team provided the panel with medical records and other evidence. On 3 February 2025, Lee presented the findings of his panel at a press conference. Lee stated that the panel "did not find any murders" and concluded that "in all cases, death or injury were due to natural causes or just bad medical care".

== Adverse effects == In clinical trials, the most common adverse effects of sildenafil use included headache, flushing, indigestion, nasal congestion, and impaired vision, including photophobia and blurred vision. Some sildenafil users have complained of seeing everything tinted blue (cyanopsia). This cyanopsia can be explained because sildenafil, while selective for PDE5, does have some affinity for PDE6, which is the phosphodiesterase found in the retina. Patients thus taking the drug may experience colorvision abnormalities. Some complained of blurriness and loss of peripheral vision. In July 2005, the US Food and Drug Administration (FDA) updated labeling for tadalafil (Cialis), vardenafil (Levitra), and sildenafil (Viagra) to reflect a small number of post-marketing reports of sudden vision loss, while acknowledging that "...it is not possible to determine whether these oral medicines for erectile dysfunction were the cause of the loss of eyesight or whether the problem is related to other factors such as high blood pressure or diabetes, or to a combination of these problems." A careful review of pooled data from clinical trials containing well documented information about the dose and duration of exposure to the drug for a large number of patients, yields no evidence for an increased risk of non-arteritic anterior ischemic optic neuropathy or other adverse ocular events associated with PDE5 inhibitor use.

The melanocortins are a family of neuropeptide hormones which are the ligands of the melanocortin receptors. The melanocortin system consists of melanocortin receptors, ligands, and accessory proteins. The genes of the melanocortin system are found in chordates. Melanocortins were originally named so because their earliest known function was in melanogenesis. It is now known that the melanocortin system regulates diverse functions throughout the body, including inflammatory response, fibrosis, melanogenesis, steroidogenesis, energy homeostasis, sexual function, and exocrine gland function. There are four endogenous melanocortin agonists which are derived from post-transcriptional processing of the precursor molecule proopiomelanocortin (POMC). They are adrenocorticotropic hormone (ACTH), α-melanocyte stimulating hormone (MSH), β-MSH, and γ-MSH. In addition to agonists which activate melanocortin receptors, there are two antagonists which inhibit receptor activity, agouti-signaling protein (ASIP) and agouti-related protein (AgRP).
