=== Identification === B. canis is a zoonotic organism. The bacteria are oxidase, catalase and urease positive and non-motile. Unlike haemophilus, which they resemble, they have no requirements for added X (hemin) and V (nicotinamide adenine dinucleotide) factors in cultures. Full identification is established by serology and PCR. Due to B. canis being naturally rough (lacking O-polysaccharide), smooth-antigen serology is unreliable. Modern practice uses B. canis-adapated serological assays such as RSAT/2-ME, IFAT (Immunofluorescence Antibody test) and ELISA to make full identification. B. canis is not acid-fast, but they tend to maintain their color when exposed to weak acids. This results in their red color when stained. When isolated, B. canis is always in a rough form, with hydrophobic LPS imbedded in its outer membrane. MALDI-TOF mass spectrometry with validated databases and whole-genome sequencing (WGS) are now increasingly used for definitive confirmation of B. canis and specific outbreak tracing. Colonies of Brucella can typically start to be seen after 48 hours. These colonies tend to be 0.5-1.0 mm in diameter, with a convex shape and are typically circular. Growth is often slower than other bacteria, with colonies requiring up to 72 hours for clear visualization. B. canis presents itself as a rough species, so colonies are generally non-mucoid rather than sticky, glue-like of presenting smooth strains. Their colonies are small, white to yellowish white and non-pigmented, consistent with rough Brucella.
== Genetics == Mutation in the gene CAPN3, which encodes the protein calpain-3 (CAPN3), is the cause of calpainopathy. As of 2019, more than 480 CAPN3 mutations have been reported, some of which can be associated with severe or benign disease course. Usually, the disease follows an autosomal recessive inheritance pattern, requiring both CAPN3 alleles to be mutated for disease to occur. === Scope of Computer Validation === The definition of validation above discusses production of evidence that a system will meet its specification. This definition does not refer to a computer application or a computer system but to a process. The main implications in this are that validation should cover all aspects of the process including the application, any hardware that the application uses, any interfaces to other systems, the users, training and documentation as well as the management of the system and the validation itself after the system is put into use. === Post-operative changes === One cause of Type 1 diabetes is through surgery. This is due to the destruction or intentional removal of a portion of or the entire pancreas. This decreases the number of beta-islet cells capable of producing insulin greatly, resulting in an acquired form of Type 1 diabetes known as pancreatogenic diabetes mellitus. This type of diabetes is most often seen in patients that undergo a pancreatoduodenectomy (a.k.a. Whipple procedure) or a total pancreatectomy. Patients who undergo a total pancreatectomy are medically recognized as having type 3c diabetes. This nomenclature informs medical professionals that the patient has no insulin production and requires extensive monitoring to avoid severe hyperglycemia or hypoglycemia. Hypoglycemia is significantly more worrying in these patients due to the potential for coma and even death, as hyperglycemia causes more subtle damage over a longer period of time and only affects consciousness at severe levels. Following these surgeries, patients may have reduced levels of insulin; they may also have reduced levels of glucagon, a hormone that raises blood sugar. Many of these patients require an insulin pump that constantly injects insulin to reduce their sugar levels .
