Tesamorelin is a synthetic peptide of forty-four amino acids whose sequence reproduces human growth hormone-releasing hormone. Its distinguishing feature sits at the amino terminus, where a trans-3-hexenoyl group replaces the free amine. That acylation slows cleavage by dipeptidyl peptidase IV, an enzyme that otherwise removes the first two residues and inactivates the natural hormone quickly. The modified peptide therefore persists longer in circulation while keeping the same receptor target. It is handled as a lyophilized solid and dissolved shortly before use.
Signaling begins at the GHRH receptor, a class B G protein-coupled receptor displayed on somatotroph cells of the anterior pituitary. Receptor occupancy activates Gs proteins, which raise adenylyl cyclase activity and intracellular cyclic AMP, in turn driving protein kinase A dependent pathways. The downstream output is synthesis and pulsatile secretion of growth hormone into the bloodstream. Hepatic tissue and peripheral sites respond by increasing insulin-like growth factor 1 production. Somatostatin and IGF-1 itself supply negative feedback that caps the size and duration of each secretory burst.

Once reconstituted, the peptide is handled as a solution and is less stable than the lyophilized powder. Aqueous solutions are commonly kept cold and used within a defined period. Buffer composition and pH influence degradation rates, with extremes of acidity or alkalinity accelerating hydrolysis. Preservatives may be added in multi-dose formats to limit microbial growth. Freezing and thawing of solutions is generally avoided because it can cause precipitation or loss of activity.
Identity and purity are assessed by reversed-phase high-performance liquid chromatography, which separates the peptide from related impurities. Mass spectrometry, often coupled to liquid chromatography, confirms molecular mass and detects chemical modifications. Peptide mapping and amino acid analysis can verify sequence integrity. Water content is measured by Karl Fischer titration, and residual solvents may be checked by gas chromatography. These methods together support batch-to-batch consistency and routine quality control.
Lyophilized tesamorelin is generally stored refrigerated at temperatures between 2 and 8 degrees Celsius. The solid form is comparatively stable when kept dry and protected from light. Moisture uptake can promote aggregation and degradation, so sealed containers with desiccant are common. Researchers typically avoid repeated temperature cycling, which may stress the peptide. Documentation accompanying reference materials usually specifies a shelf life under these conditions.
Tesamorelin is a synthetic analog of growth hormone-releasing hormone, a peptide hormone produced by the hypothalamus. The molecule retains the 44-amino-acid sequence of human GHRH and carries a trans-3-hexenoyl modification at its N-terminus. This modification increases resistance to enzymatic degradation and extends the peptide's functional stability relative to native GHRH. The compound is supplied as a lyophilized powder for reconstitution and subcutaneous administration in clinical settings. Its development code was TH9507, and it belongs to the GHRH analog class. It is not a growth hormone product; instead, it acts upstream to stimulate endogenous growth hormone release.
Clinical interest in tesamorelin arose from the need to address visceral adiposity in people living with HIV. Antiretroviral therapy improved survival but was associated in some patients with central fat accumulation, altered lipid profiles, and metabolic complications. This condition, often called HIV-associated lipodystrophy, involves excess visceral adipose tissue that is difficult to manage through diet and exercise alone. Investigators evaluated tesamorelin because GHRH analogs can stimulate growth hormone secretion and influence fat distribution without direct liposuction or invasive procedures.
A Phase 3 program led to regulatory approval in the United States in 2010 for reduction of excess visceral abdominal fat in adults with HIV and lipodystrophy. Subsequent studies examined effects on liver fat, muscle area, and metabolic markers, with mixed findings for some endpoints. Long-term cardiovascular outcomes and effects on mortality remain uncertain because most trials were relatively short and focused on imaging-based fat measurements. Use in populations without HIV has been studied experimentally but is not part of the approved indication.
Lyophilized tesamorelin is generally stored refrigerated at 2 to 8 degrees Celsius, protected from light and moisture. Peptides in this class are often kept frozen at minus 20 degrees Celsius for longer periods. Reconstituted solutions are typically used within a defined window because hydrolysis and oxidation proceed faster in liquid form. Container material and headspace also influence how long a preparation retains its expected profile. Specific stability figures depend on concentration and buffer composition.
Common analytical approaches include reversed-phase high-performance liquid chromatography for purity assessment and mass spectrometry for identity confirmation. Peptide mapping after enzymatic digestion can verify the expected sequence. Immunoassays may be used to measure the compound or its downstream markers, but they can cross-react with related peptides and require careful validation. Impurity profiles typically include truncated sequences, oxidized methionine residues, and residual solvents from synthesis. Each method reports a different property, so no single assay establishes overall quality.
Storage claims vary across suppliers, and published stability data for specific formulations are limited. Extrapolating from related peptides is common but not a substitute for direct measurement. For research use, documentation such as a certificate of analysis is often requested to confirm identity and purity. What constitutes an acceptable purity threshold depends on the intended application. Open questions remain about how temperature excursions during shipping affect long-term peptide integrity. Independent verification by an end user is not routinely reported.
=== Violation of California Labor Code === In 2009 and 2011, class-action lawsuits were filed by former workers, alleging that the company violated the California Labor Code, failed to pay overtime, failed to provide meal and rest periods, failed to pay employees upon termination, and violated California's Unfair Competition Law. Panera paid $5 million to settle all claims and denied any wrongdoing.
Neuroendocrinology is the branch of biology (specifically of physiology) which studies the interaction between the nervous system and the endocrine system; i.e. how the brain regulates the hormonal activity in the body. The nervous and endocrine systems often act together in a process called neuroendocrine integration, to regulate the physiological processes of the human body. Neuroendocrinology arose from the recognition that the brain, especially the hypothalamus, controls secretion of pituitary gland hormones, and has subsequently expanded to investigate numerous interconnections of the endocrine and nervous systems. The endocrine system consists of numerous glands throughout the body that produce and secrete hormones of diverse chemical structure, including peptides, steroids, and neuroamines. Collectively, hormones regulate many physiological processes. The neuroendocrine system is the mechanism by which the hypothalamus maintains homeostasis, regulating reproduction, metabolism, eating and drinking behaviour, energy utilization, osmolarity and blood pressure.
The cyanohydrins are a special class of nitriles. Classically they result from the addition of alkali metal cyanides to aldehydes in the cyanohydrin reaction. Because of the polarity of the organic carbonyl, this reaction requires no catalyst, unlike the hydrocyanation of alkenes. O-Silyl cyanohydrins are generated by the addition trimethylsilyl cyanide in the presence of a catalyst (silylcyanation). Cyanohydrins are also prepared by transcyanohydrin reactions starting, for example, with acetone cyanohydrin as a source of HCN. Cyanohydrins can also be prepared by addition of an alkali cyanide to an aldehyde or ketone in the presence of acetic acid. For less reactive substrates, diethylaluminum cyanide provides a suitable alternative. Another approach is transhydrocyanation, in which hydrogen cyanide is transferred from acetone cyanohydrin to an aldehyde or ketone. Suitable catalysts for this transformation include lanthanide alkoxides such as lanthanum(III) isopropoxide, cerium(III) isopropoxide, samarium(III) isopropoxide, and ytterbium(III) isopropoxide. Addition of trimethylsilyl cyanide to aldehydes or ketones affords cyanohydrins as their trimethylsilyl ethers. Suitable catalysts include zinc iodide, potassium cyanide in combination with 18-crown-6, or ytterbium(III) cyanide. Under appropriate conditions, such reactions can be rendered enantioselective. Vanadium- or titanium-based catalysts bearing chiral salen-type ligands are suitable, as is the combination of tetraisopropyl orthotitanate with a chiral imine.

The mechanism describing the formation of the ozone layer was described by British mathematician and geophysicist Sydney Chapman in 1930, and is known as the Chapman cycle or ozone–oxygen cycle. Molecular oxygen absorbs high energy sunlight in the UV-C region, at wavelengths shorter than about 240 nm. Radicals produced from the homolytically split oxygen molecules combine with molecular oxygen to form ozone. Ozone in turn is photolyzed much more rapidly than molecular oxygen as it has a stronger absorption that occurs at longer wavelengths, where the solar emission is more intense. Ozone (O3) photolysis produces O and O2. The oxygen atom product combines with atmospheric molecular oxygen to reform O3, releasing heat. The rapid photolysis and reformation of ozone heat the stratosphere, resulting in a temperature inversion. This increase of temperature with altitude is characteristic of the stratosphere; its resistance to vertical mixing means that it is stratified. Within the stratosphere temperatures increase with altitude (see temperature inversion); the top of the stratosphere has a temperature of about 270 K (−3°C or 26.6°F). This vertical stratification, with warmer layers above and cooler layers below, makes the stratosphere dynamically stable: there is no regular convection and associated turbulence in this part of the atmosphere. However, exceptionally energetic convection processes, such as volcanic eruption columns and overshooting tops in severe supercell thunderstorms, may carry convection into the stratosphere on a very local and temporary basis.
=== Adipose tissue === Adipose tissue, commonly known as fat, is a depository for energy in order to conserve metabolic homeostasis. As the body takes in energy in the form of glucose, some is expended, and the rest is stored as glycogen (primarily in the liver, muscle cells), or as triglyceride in adipose tissue. An imbalance in glucose intake and energy expenditure has been shown to lead to both adipose cell hypertrophy and hyperplasia, which lead to obesity. In addition, mutations in GLUT4 genes in adipocytes can also lead to increased GLUT4 expression in adipose cells, which allows for increased glucose uptake and therefore more fat stored. If GLUT4 is over-expressed, it can actually alter nutrient distribution and send excess glucose into adipose tissue, leading to increased adipose tissue mass.
=== Nuclear actin === Nuclear actin was first noticed and described in 1977 by Clark and Merriam. Authors describe a protein present in the nuclear fraction, obtained from Xenopus laevis oocytes, which shows the same features as skeletal muscle actin. Since that time there have been many scientific reports about the structure and functions of actin in the nucleus. The controlled level of actin in the nucleus, its interaction with actin-binding proteins (ABP) and the presence of different isoforms allows actin to play an important role in many important nuclear processes.
== Structure == AgRP is a paracrine signaling molecule made of 112 amino acids (the gene product of 132 amino acids is processed by removal of the N-terminal 20-residue signal peptide domain). It was independently identified by two teams in 1997 based on its sequence similarity with agouti signalling peptide (ASIP), a protein synthesized in the skin controlling coat colour. AgRP is approximately 25% identical to ASIP. The murine homologue of AgRP consists of 111 amino acids (precursor is 131 amino acids) and shares 81% amino acid identity with the human protein. Biochemical studies indicate AgRP to be very stable to thermal denaturation and acid degradation. Its secondary structure consists mainly of random coils and β-sheets that fold into an inhibitor cystine knot motif. AGRP maps to human chromosome 16q22 and Agrp to mouse chromosome 8D1-D2.

Folgers is promoted with the slogan "The best part of waking up is Folgers in your cup!" It is well associated with a jingle featured in almost every advertisement since 1984, with lyrics by Susan Spiegel Solovay and Bill Vernick, and music by Leslie Pearl. Over the years it has been rearranged and performed by many famous musicians, such as Richie Havens, Randy Travis, Bonnie Tyler, Paul Stanley, Aretha Franklin, and Rockapella. From 1965 to 1986, Folgers was known for television ads involving "Mrs. Olson", a Swedish neighbor played by Virginia Christine who invariably recommended a cup of Folgers coffee for the characters in the commercial. Folgers promoted their instant coffee in the 1970s and early 1980s ads which took the viewer inside various 'high-end' restaurants while a voice-over (by Bryan Clark) whispered to the viewer that they've secretly switched the coffee used at the restaurant with Folgers, and watched the restaurants' patrons to see if they could tell whether or not they noticed the difference. One Folgers television ad from 1985, "Peter Comes Home For Christmas," became particularly associated with the Christmas holidays. A college student returns home, apparently on Christmas Day. His younger sister is the first one to greet Peter, helping him make the Folgers. The smell of freshly brewed coffee awakening his parents and alerting them to their son's arrival. The Cunningham & Walsh spot aired yearly until 1998, then in edited form in 2004 and 2005. In the mid-1980s Folgers became a NASCAR sponsor.
== History == The pancreas was first identified by Herophilus (335–280 BC), a Greek anatomist and surgeon. A few hundred years later, Rufus of Ephesus, another Greek anatomist, gave the pancreas its name. Etymologically, the term "pancreas", a modern Latin adaptation of Greek πάγκρεας, [πᾶν ("all", "whole"), and κρέας ("flesh")], originally means sweetbread, although literally meaning all-flesh, presumably because of its fleshy consistency. It was only in 1889 when Oskar Minkowski discovered that removing the pancreas from a dog caused it to become diabetic. Insulin was later isolated from pancreatic islets by Frederick Banting and Charles Best in 1921. The way the pancreatic tissue has been viewed has also changed. Previously, it was viewed using simple staining methods such as H&E stains. Now, immunohistochemistry can be used to differentiate cell types more easily. This involves visible antibodies to the products of certain cell types, and helps identify with greater ease cell types such as alpha and beta cells.
=== Dendritogenesis === Local interaction of BDNF with the TrkB receptor on a single dendritic segment is able to stimulate an increase in PSD-95 trafficking to other separate dendrites as well as to the synapses of locally stimulated neurons. PSD-95 localizes the actin-remodeling GTPases, Rac and Rho, to synapses through the binding of its PDZ domain to kalirin, increasing the number and size of spines. Thus, BDNF-induced trafficking of PSD-95 to dendrites stimulates actin remodeling and causes dendritic growth in response to BDNF.

The amino acid sequence matches human growth hormone-releasing hormone, but the amino terminus carries a trans-3-hexenoyl group instead of a free amine. That single structural change chiefly affects enzymatic stability rather than receptor selectivity.
