Everything below concerns somatotroph. We keep the language plain, cite what the science says, and separate well-supported claims from open questions.
Updated 2026-07-14. Numbers and descriptions here follow the published literature rather than marketing material.
Signaling begins at the GHRH receptor, a class B G protein-coupled receptor displayed on somatotroph cells of the anterior pituitary. Receptor occupancy activates Gs proteins, which raise adenylyl cyclase activity and intracellular cyclic AMP, in turn driving protein kinase A dependent pathways. The downstream output is synthesis and pulsatile secretion of growth hormone into the bloodstream. Hepatic tissue and peripheral sites respond by increasing insulin-like growth factor 1 production. Somatostatin and IGF-1 itself supply negative feedback that caps the size and duration of each secretory burst.
Metabolic interest in this compound centers on fat distribution rather than on hormone levels alone. Imaging trials in adults with excess abdominal fat report reductions in visceral adipose tissue, while subcutaneous depots change comparatively little. Growth hormone and IGF-1 are presumed to carry the effect, but the separate contribution of each is not firmly established. Whether these changes persist after treatment stops, and whether they alter longer-term health outcomes, remain open questions that published work does not answer consistently.

Several related peptides act on the same receptor, including sermorelin, a shorter GHRH fragment, and modified analogs such as CJC-1295 and modified GRF(1-29) that are common in research settings rather than approved products. Tesamorelin differs from growth hormone itself in that it acts upstream, prompting the pituitary to release the hormone through physiological signaling rather than supplying it directly. Terminology in the literature distinguishes GHRH analogs, growth hormone secretagogues, and recombinant growth hormone, although popular discussion often blurs these categories together. Precise naming matters when comparing study results.
Tesamorelin is a synthetic peptide of 44 amino acids that reproduces the sequence of human growth hormone-releasing hormone (GHRH) and carries a trans-3-hexenoyl group on its N-terminal tyrosine. That small fatty-acid modification blocks cleavage by dipeptidyl peptidase-4, the enzyme that rapidly degrades native GHRH in plasma. The result is a molecule with a longer circulating half-life than the natural hormone while retaining the same receptor target. It is supplied as a lyophilized powder for reconstitution and belongs to the broader class of GHRH analogs studied for effects on pituitary growth hormone secretion.
== The document == The document was presented to the British Museum in 1924 by Walter Rothschild; today it is held in the British Library, which separated from the British Museum in 1973, as Additional Manuscripts number 41178. From October 1987 to May 1988 it was lent outside the UK for display in Israel's Knesset.
=== Metal sulfides === Sulfur reacts with many metals. Electropositive metals give polysulfide salts. Copper, zinc, and silver are attacked by sulfur; see tarnishing. Although many metal sulfides are known, most are prepared by high temperature reactions of the elements. Geoscientists also study the isotopes of metal sulfides in rocks and sediment to study environmental conditions in the Earth's past.
In April 2002, Yosri Fouda of Al Jazeera met al-Qaeda member Khalid Sheikh Mohammed and bin al-Shibh, who were in hiding, and they admitted to him their involvement in the attacks. Mohammed had been an organizer and financier of al-Qaeda's 1993 bombing of the World Trade Center, and he was the uncle of Ramzi Yousef, the lead bomber in that attack. Mohammed and Yousef planned another terrorist attack, the Bojinka plot, for January 1995. Despite its failure, and Yousef's imprisonment by the U.S. afterwards, the plot would influence the later September 11 attacks. The 2004 9/11 Commission Report determined that the animosity that Mohammed, the principal architect of 9/11, felt towards the U.S. had stemmed from his "violent disagreement with U.S. foreign policy favoring Israel."
In Table 5, the approximately $85 billion fine chemical market is subdivided into major applications according to their relevance: fine chemicals for pharmaceuticals, agrochemicals, and specialty chemicals outside life sciences. Furthermore, a distinction is made between captive (in-house) production and merchant market. Pharmaceutical fine chemicals (PFCs) account for two-thirds of the total. Out of the PFC value of $55 billion, about $23 billion (~40%) are traded, and $32 billion (~60%) are the production value of the pharma industry's in-house production. Within life science products, fine chemicals for agro and veterinary drugs follow in importance. The production value for fine chemicals used for specialty chemicals other than pharmaceuticals and agrochemicals is estimated at $15 billion. As the leading specialty chemical companies (Akzo Nobel, Dow, DuPont, Evonik, Chemtura and Mitsubishi) are backward-integrated, the share of in-house production is estimated at 75%, leaving a merchant market of approximately $5 billion.
Clobazam is predominantly a positive allosteric modulator at the GABAA receptor to increase GABAergic transmission, particularly chloride conductance in neurons and with some speculated additional activity at sodium channels and voltage-sensitive calcium channels. Clobazam binds at a distinct binding site associated with a Cl− ionophore at the GABAA receptor, increasing the duration of time for which the Cl− ionophore is open. The post-synaptic inhibitory effect of GABA in the thalamus is prolonged as a result. The exact mechanism of action for clobazam, a 1,5-benzodiazepine, which has anxiolytic and anticonvulsant effects similar to those produced by other benzodiazepine derivatives. Clobazam is a potent benzodiazepine receptor partial agonist at the GABAA receptor and the effects are related to binding to one or more specific GABA receptor subunits, increasing GABA-mediated inhibition. Clobazam is thought to involve the potentiation of GABAergic neurotransmission resulting from binding at the benzodiazepine site of the GABAA receptor. Like other 1,5-benzodiazepines (for example, arfendazam, lofendazam, triflubazam, and CP-1414S), clobazam and the active metabolite N-desmethylclobazam have less affinity for the α1 subunit (sedative effects) of the GABAA receptor compared to the 1,4-benzodiazepines. They have a higher affinity for the α2 subunit (anxiolytic effects) and γ2 subunit of the GABAA receptor, which is essential for the anxiolytic and anticonvulsant effects of clobazam.

=== Drug target === The anticonvulsant gabapentin [Neurontin; 1-(aminomethyl)cyclohexaneacetic acid] is a drug often used to treat patients with neuropathic pain. This neuropathic pain can be caused by a number of things, including diabetic neuropathy and postherpetic neuralgia. Gabapentin is an amino acid drug structurally similar to the two neurotransmitters glutamate (synthesized by BCATs) and GABA. The drug competitively inhibits both BCAT isoforms in the brain, slowing down glutamate production. Gabapentin also inhibits GABA aminotransferase (GABA-T) and glutamate dehydrogenase (GDH), two other enzymes in the glutamate and GABA metabolic pathway.
== Pathophysiology == Coronary occlusion is caused by plaque inside of the blood vessels that direct oxygen rich blood to the heart. Plaque is caused by fatty deposits and scar tissue that cling to the walls of coronary arteries. The development of plaque takes years and leads to stenosis of the coronary arteries and progressively reduces blood flow. Due to the slower development of this condition, the body will adapt and create small blood vessels that circumvent the blockage. The small blood vessels form a natural bypass of the blockage, but often do not supply enough blood to meet an increased demand when stressors are applied like exercise. When a plaque has a greater than 50% diameter stenosis, the reduced blood flow through the coronary artery during exertion may lead to angina. Acute coronary events occur when a thrombus forms due to disruption of a plaque. In acute heart attack, occlusion is greater than in unstable angina, where arterial occlusion is not full blockage. Downstream embolism of thrombus may also produce microinfarcts.
== Structure == Relaxin-3 consists of 51 amino acids in humans which are arranged into a two-chain structure (designated the A-chain and B-chain). There are three disulfide bonds (two interchain, one intrachain), with the residues that mediate binding to/activation of RXFP3 residing within the B-chain. At translation, pro-relaxin-3 also contains a C-chain (in between the A and B-chains) which is removed via protolytic cleavage to form the mature neuropeptide.
==== Natural casing ==== As with most sausages, hot dogs must be in a casing to be cooked. Traditional casing is made from the small intestines of sheep. The products are known as "natural casing" hot dogs or frankfurters. These hot dogs have firmer texture and a "snap" that releases juices and flavor when the product is bitten. Kosher casings are expensive in commercial quantities in the US, so kosher hot dogs are usually skinless or made with reconstituted collagen casings.

== History == The precise origin of absinthe is unclear. The medical use of wormwood dates back to ancient Egypt and is mentioned in the Ebers Papyrus from around 1550 BC. Wormwood extracts and wine-soaked wormwood leaves were used as remedies by the ancient Greeks. Moreover, some evidence exists of a wormwood-flavoured wine in ancient Greece called absinthites oinos. Lucretius' De Rerum Natura (I, 936–950) says that a drink containing wormwood was given as medicine to children in a cup with honey on the brim to make it drinkable. The first evidence of absinthe, in the sense of a distilled spirit containing green anise and fennel, dates to the 18th century. According to popular legend, it began as an all-purpose patent remedy created by Dr. Pierre Ordinaire, a French doctor living in Couvet, Switzerland, around 1792 (the exact date varies by account). Ordinaire's recipe was passed on to the Henriod sisters of Couvet, who sold it as a medicinal elixir. By other accounts, the Henriod sisters may have been making the elixir before Ordinaire's arrival. In either case, a certain Major Dubied acquired the formula from the sisters in 1797 and opened the first absinthe distillery named Dubied Père et Fils in Couvet with his son Marcellin and son-in-law Henry-Louis Pernod. In 1805, they built a second distillery in Pontarlier, France, under the company name Maison Pernod Fils. Pernod Fils remained one of the most popular brands of absinthe until the drink was banned in France in 1914.
== History == Before amylin deposition was associated with diabetes, already in 1901, scientists described the phenomenon of "islet hyalinization", which could be found in some cases of diabetes. A thorough study of this phenomenon was possible much later. In 1986, the isolation of an aggregate from an insulin-producing tumor was successful, a protein called IAP (Insulinoma Amyloid Peptide) was characterized, and amyloids were isolated from the pancreas of a diabetic patient, but the isolated material was not sufficient for full characterization. This was achieved only a year later by two research teams whose research was a continuation of the work from 1986.
Oclacitinib, sold under the brand name Apoquel among others, is a veterinary medication used in the control of atopic dermatitis and pruritus from allergic dermatitis in dogs at least 12 months of age. Chemically, it is a synthetic cyclohexylamino-pyrrolopyrimidine Janus kinase inhibitor (JAK inhibitor) that is relatively selective for JAK1. It inhibits signal transduction when the JAK is activated and thereby helps downregulate expression of inflammatory cytokine. Oclacitinib was approved for use in the United States in 2013 and in the European Union in 2023. A flavored chewable form was approved for use in the United States in 2023.
== Aging studies == A gradual increase in CD38 has been implicated in the decline of NAD+ with age. Treatment of old mice with a specific CD38 inhibitor, 78c, prevents age-related NAD+ decline. CD38 knockout mice have twice the levels of NAD+ and are resistant to age-associated NAD+ decline, with dramatically increased NAD+ levels in major organs (liver, muscle, brain, and heart). On the other hand, mice overexpressing CD38 exhibit reduced NAD+ and mitochondrial dysfunction. Macrophages are believed to be primarily responsible for the age-related increase in CD38 expression and NAD+ decline. Cellular senescence of macrophages increases CD38 expression. Macrophages accumulate in visceral fat and other tissues with age, leading to chronic inflammation. The inflammatory transcription factor NF-κB and CD38 are mutually activating. Secretions from senescent cells induce high levels of expression of CD38 on macrophages, which becomes the major cause of NAD+ depletion with age. Decline of NAD+ in the brain with age may be due to increased CD38 on astrocytes and microglia, leading to neuroinflammation and neurodegeneration.
The Australian Rugby League Commission Limited (ARL), formerly the Australian Rugby Football League Limited is an Australian rugby league football competition operator. It was founded in 1986 as the Australian Rugby Football League Limited and succeeded the Australian Rugby Football League Board of Control which had been formed in 1924. Since its inception, the ARL has administered the Australian national team and represented Australia in international rugby league matters. Prior to 1998, the code in Australia had been principally administered by individual state leagues on a domestic basis, and the ARL on a national and international basis.

The amino acid sequence matches human growth hormone-releasing hormone, but the amino terminus carries a trans-3-hexenoyl group instead of a free amine. That single structural change chiefly affects enzymatic stability rather than receptor selectivity.
Growth hormone released from the pituitary stimulates IGF-1 production in the liver and other tissues, so circulating IGF-1 generally rises during exposure. The size of the rise varies between individuals and depends on baseline hormonal status and other concurrent factors.
Reported imaging studies focus on visceral adipose tissue, where reductions are more consistently observed across trials. Subcutaneous depots show smaller and less reproducible changes, so the two compartments should not be treated as equivalent.
tesamorelin 属 GHRH 类似物,作用于垂体受体以促进内源生长激素释放;生长激素本身是直接补充的外源激素。两者在给药逻辑、作用位点和反馈调控路径上并不相同。
